Reference standards for LC-MS/MS
LC-MS/MS quantifies a compound by its retention time and two mass transitions. Because the detector responds to the analyte itself, the calibration curve is only as good as the reference material behind it — an impurity that co-elutes, or a standard whose assigned content is wrong, shifts every result in the run.
What this method asks of a standard
| Requirement | Why it matters |
|---|---|
| An assigned content, not just a purity | A chromatographic purity figure cannot be used as a gravimetric standard. LC-MS/MS calibration needs the mass fraction of the analyte in the material, which is what a certificate of analysis states per lot. |
| Isotope-labelled internal standard | A stable-isotope analogue co-elutes with the analyte and ionises similarly, which compensates for matrix effects and extraction losses. Deuterated and 13C-labelled analogues are the usual choice; the label should sit where it cannot exchange. |
| Metabolite and degradation products | Metabolites and forced-degradation products are the analytes that methods for impurity profiling and stability studies must be calibrated against. |
| Nitrosamine standards | Nitrosamine and NDSRI methods are LC-MS/MS methods in most laboratories; they need the specific nitrosamine, not a parent-drug standard. |
Standard types used with LC-MS/MS
These are the catalogue groups most often quoted for LC-MS/MS work. They are entry points into the catalogue, not a statement that any particular lot suits any particular method — that is established in your own validation, against the certificate for the lot.
