Codeine impurities and related standards

Codeine sulfate is an opioid agonist with a relatively weak affinity for the mu-opioid receptor, and the label attributes most of its analgesia to conversion to morphine in the body rather than to the parent alkaloid. Tablets are used for mild to moderately severe pain, and codeine has also been prescribed as an antitussive. The salt choice is a real analytical variable: the sulfate and the phosphate differ in mass, solubility and water content, the monographs are written separately for each, and the label instructs dispensing in a tight, light-resistant container. Related-substance work follows the chemistry of the morphinan skeleton. The 3-methoxy group is what distinguishes codeine from morphine, so morphine is the obvious process residue and the first related substance to look for in a methylation route, while the tertiary amine generates N-demethylated and N-oxide material under oxidative stress. The C6 alcohol is secondary and sits next to the 7,8 double bond, which makes it the part of the molecule most exposed to oxidation and to photolysis. Identity by NMR and MS, a declared content basis, and the specified-impurity list of the monograph are the buyer's checks.

Frequently asked

How many Codeine-related reference standards do you hold?
41 standards are listed on this page as of the latest catalogue build. CAS numbers are published for 41 of them and 41 entries carry a structure drawing. The catalogue is extended as new lots are released.
Can you supply a Codeine impurity that is not listed here?
Yes. Non-compendial impurities can be quoted from a structure — a CAS number is not required. Send the structure, a SMILES string or the parent drug substance with your enquiry and we will come back with availability.
What documentation is supplied with Codeine impurities?
Every lot ships with its own certificate of analysis. The certificate records the catalogue number, the batch number, the tests performed, the specification applied and the result obtained for that lot. A safety data sheet is supplied with the shipment.
Do you publish prices or a lead time for Codeine impurities?
No price list is published: quotations are prepared per enquiry, because availability and packing depend on the lot and the quantity you need. Send the catalogue number and the quantity and a technical contact will answer with availability and a quotation.
